Hi,
I just installed 4.2.3 with the Windows self-installer. This works fine.
Now I wanted to upgrade to 4.2.4. So I downloaded the Upgrade package and replaced the old files with the new ones. Now I get the message when I want to go to the start page: Internal Server Error (500), the Server encountered an internal error or misconfiguration and was unable to complete you request.
How to make the upgrade correct?

Check the Apache error logs for details. I do not recommend to use the Windows installer, the developer never open sourced the installer and he left the TWiki community. Do a native install, or a "TWiki on Linux in VMware in Windows" install.

Itīs a pity that you donīt recommend the Windows installer. This is definitely the easiest to get a TWiki working!
There are as well installations "TWIki on Linux VMware in Windows" from the same developer as the Windows installer.

You could try the somewhat outdated
TWikiVMDebianStable.

If you use "TWiki on Linux in VMware in Windows" can it then been seen on the intranet?
I have to say that I would like it to been seen. For testing purposes.

That depends on your firewall settings and on how your VMware image is configured to connect to the network.
